This project will develop an innovative fluorescent biosensor for investigating the spatiotemporal dynamics of insulin within the brain. While the mechanisms governing insulin transport into the brain are relatively well characterized, its diffusion and distribution within the cerebral extracellular space and matrix remain poorly understood, despite their potential involvement in metabolic and neurodegenerative disorders. The proposed strategy relies on the synthesis of polymeric nanoparticles functionalized with insulin-specific aptamers, enabling the optical detection of insulin through Förster resonance energy transfer (FRET). Following comprehensive physicochemical characterization and ex vivo validation, the biosensor will be evaluated in vivo at the single-cell level using advanced fluorescence microscopy approaches. This novel tool will provide insights into the cerebral bioavailability, distribution, and signaling of insulin, thereby contributing to a better understanding of its role in metabolic and neurodegenerative diseases and potentially opening new avenues for investigating their underlying mechanisms.
